ABSTRACT

Objective To study the changes in serum homocysteine (Hcy) and matrix metalloproteinase-9 (MMP-9) levels and risk factors in patients with coronary heart disease (CHD) complicated with Helicobacter pylori (HP) infection in Chengdu area, and to provide a theoretical basis for the prevention of HP infection in patients with coronary heart disease. Methods A total of 348 CHD patients admitted to our hospital in Chengdu from 2019 to 2021 were selected. Hp infection status was detected by C14 urea breath test. Patients were classified into control group (n=197) and HP infection group (n=151) according to the detection results. Data including gender, age, body mass index and peptic ulcer history were collected, and univariate analysis and logistic regression were used to screen the risk factors affecting the occurrence of HP infection in patients with CHD. Results The prevalence rate of HP infection was 43.39% (151/348) among the selected CHD patients. Serum levels of Hcy and MMP-9 were notably elevated in HP infection group compared with control group (P<0.05). The proportion of patients with age ≥60 years old, hyperlipidemia, proton pump inhibitor use history, and frequent consumption of out-of-home food and spicy food in HP infection group was obviously larger than that in control group (P<0.05). Hyperlipidemia (OR=3.719), history of proton pump inhibitor use (OR=3.254) and frequent consumption of out-of-home food (OR=2.721) were independent risk factors for HP infection in CHD patients (P<0.05). Conclusion CHD patients in Chengdu suffer a prevalence rate of HP infection, and have elevated levels of serum Hcy and MMP-9. Furthermore, the intervention measures for patients with hyperlipidemia, proton pump inhibitor drug use history and frequent consumption of out-of-home food are of vital importance for decreasing the risk of HP infection.

ABSTRACT

Objective:To investigate the impact of trigger timing of gonadotropin- releasing hormone (GnRH) antagonist regimen for infertility patients of various ages. Methods:This was a retrospective study, 1 529 infertility patients who receiving GnRH antagonist regimen in Chongqing Health Center for Women and Children from January 2017 to December 2018 were divided into the advance trigger group and the standard trigger group, and further divided into three subgroups according to age:<35 years, 35-40 years,>40 years. The number of retrieved oocytes and transplantable embryos, the clinical pregnancy rate and the live birth rate among patients in the advance trigger group and standard trigger group in various age subgroups were compared.Results:(1) The gonadotropin (Gn) days among the three age subgroups were significantly shorter in the advance trigger group compared to the same-aged standard trigger group (all P<0.01), but only in the 35-40 years and >40 years subgroups, the Gn doses in the advance trigger group [(2 702±551) and (2 780±561) U] were significantly less than those in the standard trigger group (all P<0.01). In the <35 years subgroup, the number of oocytes retrieved and transplantable embryos of the advance trigger group (6.6±4.8 and 2.6±2.7) were significantly less than those of the standard trigger group (all P<0.01), but there was no difference in the number of top-quality embryos ( P=0.580); however, in the 35-40 years and >40 years subgroups, there were no significant differences between advance and standard trigger groups in terms of the afore mentioned 3 indicators (all P>0.05), only the numbers of top-quality embryos in the advance trigger group (0.6±1.0 and 0.6±0.9) were significantly higher than those in the standard trigger group (all P<0.01). (2) In the <35 years and 35-40 years subgroups, no significant differences were noted between the advance trigger group and standard trigger group with regard to the clinical pregnancy rate and live birth rate (all P>0.05); but in the >40 years subgroup, the clinical pregnancy rate of the advance trigger group was significantly higher than that of the standard trigger group [33.0% (30/91) vs 19.2% (25/130), P=0.020], and there was no statistical difference in the live birth rate ( P=0.064). (3) Multivariate logistic regression analysis showed that trigger timing was an independent predictor of clinical pregnancy rate in the >40 years subgroup ( OR=0.334, 95% CI: 0.119-0.937, P=0.037), but not an independent predictor of live birth rate ( P>0.05). Conclusions:Advance trigger in the GnRH antagonist protocol for infertility patients >40 years old could effectively reduce Gn times and Gn dosage, increase the number of top-quality embryos, and improve the clinical pregnancy rate. Therefore, compared with patients ≤40 years of age, patients >40 years might benefit more from advance trigger.

ABSTRACT

Objective@#To explore the application of new pretreatment technology of automatic electrothermal digestion instrument in the detection of arsenic in urine samples, in order to achieve rapid, convenient and accurate detection results.@*Methods@#Using wet digestion, an advanced sample preparation and digestion method (referred to as new pretreatment technology) was established by hydride generation-atomic fluorescence spectrometry (HG-AFS). Wet digestion: took 1.0 ml urine samples and put them into 10.0 ml corked scale digestive tubes, and added 0.20 ml sulfuric acid, 0.30 ml nitric acid and 0.50 ml hydrogen peroxide into the tubes successively. Shook solution and put them at AED-IV automatic electrothermal digestion instrument with 160 ℃ for half an hour, then heated up to 200 ℃ for 1 hour. It needed to be continually heated up to 260 ℃ and dissolved the solution until colorless transparent or light yellow. When the liquid level calm, and uniform reflux appeared, which meant the digestion finished, and then measured by atomic fluorescence photometer. At the same time, the new pretreatment technology was compared with the current standard (WS/T 474-2015) method, and the standard curve and linear range were calculated, as well as the detection line, precision and accuracy of the sample measurement.@*Results@#The new pretreatment technology could digest 60 samples per sample, which was superior to the current standard methods (20 samples) and reduced the amount of reagent used. There was no statistically significant difference between the two methods (P > 0.05). In the linear range of 0-100 μg/L, the correlation coefficient (r) > 0.999 3, the detection limit of the method was 0.066 μg/L, and the relative standard deviation (RSD) < 5%.@*Conclusions@#The new pretreatment technology is fast, efficient, environmentally friendly, economical, highly automated. The experimental result error is in a controllable range. It can be used for digestion of arsenic content in urine of different concentrations and has high applicability.

ABSTRACT

Objective To detect the methylation status of SALL3 gene promoter region in normal cervical tissues, cervical cancer tissues, and cervical cancer cell lines and thus explore the relationship between methylation status and the expression of SALL3 gene. Methods The DNA methylation statuses of SALL3 gene in normal cervical, cervical cancer tissues and cervical cancer cell lines were analyzed by methylation-specific PCR(MS-PCR).The expressions of SALL3 mRNA in cervical cancer cell lines, cervical cancer tissues, and normal cervical tissues were detected by RT-PCR.Results In cervical cancer and matched peri-carcinomatous samples, the methylation levels of SALL3 were up-regulated(CCa .CCap:=0.046;CCa .NC =0.039)and the protein expressions were down-regulated(CCa .CCap:=0.012;CCa .NC =0.000)when compared with normal cervix samples. The mRNA levels of SALL3 in HeLa and SiHa cells treated with 5-Azacytidine were elevated in a dose-dependent manner(HeLa:=0.001;SiHa:=0.002).The methylation level of SALL3 was higher in high risk human papillomavirus(HPV)-positive cervical samples than in HPV-negative cervical samples(=0.014),which also resulted in a descending SALL3 expression in HPV-positive samples(=0.021).Conclusions The hypermethylation of SALL3 in promoter regions inhibits the expression of SALL3 in cervical cancer tissue samples.Infection with high-risk HPV serotypes may increase the methylation of SALL3 promoter region, silence its expression, and thus promote the development of cervical cancer.

ABSTRACT

OBJECTIVETo investigate the potential efficacy of panaxadiol saponins component (PDS-C), a biologically active fraction isolated from total ginsenosides, to reverse chemotherapy-induced myelosuppression and pancytopenia caused by cyclophamide (CTX).

METHODSMice with myelosuppression induced by CTX were treated with PDS-C at a low- (20 mg/kg), moderate- (40 mg/kg), or high-dose (80 mg/kg) for 7 consecutive days. The level of peripheral white blood cell (WBC), neutrophil (NEU) and platelet (PLT) were measured, the histopathology and colony formation were observed, the protein kinase and transcription factors in hematopoietic cells were determined by immunohistochemical staining and Western blot.

RESULTSIn response to PDS-C therapy, the peripheral WBC, NEU and PLT counts of CTX-induced myelosuppressed mice were significantly increased in a dose-dependent manner. Similarly, bone marrow histopathology examination showed reversal of CTX-induced myelosuppression with increase in overall bone marrow cellularity and the number of hematopoietic cells (P<0.01). PDS-C also promoted proliferation of granulocytic and megakaryocyte progenitor cells in CTX-treated mice, as evidenced by significantly increase in colony formation units-granulocytes/monocytes and -megakaryocytes (P<0.01). The enhancement of hematopoiesis by PDS-C appears to be mediated by an intracellular signaling pathway, this was evidenced by the up-regulation of phosphorylated mitogen-activated protein kinase (p-MEK) and extracellular signal-regulated kinases (p-ERK), and receptor tyrosine kinase (C-kit) and globin transcription factor 1 (GATA-1) in hematopoietic cells of CTX-treated mice (P<0.05).

CONCLUSIONSPDS-C possesses hematopoietic growth factor-like activities that promote proliferation and also possibly differentiation of hematopoietic progenitor cells in myelosuppressed mice, probably mediated by a mechanism involving MEK and ERK protein kinases, and C-kit and GATA-1 transcription factors. PDS-C may potentially be a novel treatment of myelosuppression and pancytopenia caused by chemotherapy.

ABSTRACT

Three kinds of glass sharp electrospray ionization devices were built for direct mass spectrometry (MS) analysis using sharp slide, slender capillary and sharp dropper. Through the simulation experiment and mass spectrometry experimental investigation, the influence of the experimental conditions was studied. Under the optimum working conditions such as the electrospray tip diameter was less than 0.24 mm, the distance between the electrospray tip and the MS sample inlet was 10 mm, the electrospray voltage was 4.5 kV, the electrospray capillary angle was about 5.0°-7.5° (upper at back) and the left and right angles were 0°, 0.1 mg/mL sample solution(containing 1% formic acid for positive ion detection,or ammonium hydroxide for negative ion detection) could be well analyzed by MS. In positive or negative ion modes, the quinine, mandelic acid, benzoin, phenylalanine and other different types of sample solutions were analyzed by MS. The result showed that quinine, mandelic acid, benzoin, phenylalanine, tartaric acid,2,5-dihydroxybenzoic acid (DHB) and tangeretin could be detected in positive ion mode using sharp dropper electrospray. DHB, phenylalanine, tartaric acid and mandelic acid were detected in negative ion mode. Sharp slide electrospray was suitable for measuring the stable sample in the air, capillary electrospray and sharp dropper electrospray were suitable for detecting many low viscosity sample solution even instable sample. Using the method, the extraction solution of propranolol hydrochloride tablet and berberine hydrochloride tablet was directly analyzed by MS. The results showed that the glass sharp electrospray method had the advantages such as without carrier gas, simple structure, easy to build and operate, rapid analysis, anti-cross pollution, and could be conveniently used for MS analysis.

ABSTRACT

Cell transdifferentiation is the direct conversion of one type of cell into another without becoming a pluripotent stem cell. Rapid progress in this field has brought new perspectives on understanding and treating disorders associated with the nervous system, such as neurodegeneration, traumatic injuries, and neuropsychiatric diseases.Being able to obtain functional neurons through trans-differentiation from abundant somatic cells also provides a new strategy for regenerative medicine. In this review we highlight the recent progress in cell transdifferentiation for neurological disorders and dis-cuss potential challenges facing its clinical applications.

ABSTRACT

OBJECTIVETo explore the effects of bufalin on inhibiting proliferation, up-regulating methylation of Wilm' tumor 1 gene (WT1) as well as its possible mechanisms in human erythroid leukemic (HEL) cells.

METHODSThe HEL cells were treated with bufalin at various concentrations to observe cellular morphology, proliferation assay and cell cycle. The mRNA and protein expression levels of WT1 were detected by reverse transcription polymerase chain reaction (RT-PCR), Western blot and immunocytochemistry, DNA methylation of WT1 and protein expression levels of DNA methyltransferase 3a (DNMT3a) and DNMT3b were analyzed by methylation-specific PCR, and Western blot respectively.

RESULTSThe bufalin was effective to inhibit proliferation of HEL cells in a dose-dependent manner, their suppression rates were from 23.4%±2.1% to 87.2%±5.4% with an half maximal inhibit concentration (IC) of 0.046 μmol/L. Typical apoptosis morphology was observed in bufalin-treated HEL cells. The proliferation index of cell cycle decreased from 76.4%±1.9% to 49.7%±1.3%. The expression levels of WT1 mRNA and its protein reduced gradually with increasing doses of bufalin, meanwhile, the methylation status of WT1 gene changed from unmethylated into partially or totally methylated. While, the expression levels of DNMT3a and DNMT3b protein gradually increased by bufalin treatment in a dose-dependent manner.

CONCLUSIONSBufalin can not only significantly inhibit the proliferation of HEL cells and arrest cell cycle at G/Gphase, but also induce cellular apoptosis and down-regulate the expression level of WT1. Our results provide the evidence of bufalin for anti-leukemia, its mechanism may involve in increasing WT1 methylation status which is related to the up-regulation of DNMT3a and DNMT3b proteins in erythroid leukemic HEL cells.

ABSTRACT

BACKGROUNDThe emergency department (ED) has a pivotal influence on the management of acute heart failure (AHF), but data concerning current ED management are scarce. This Beijing AHF Registry Study investigated the characteristics, ED management, and short- and long-term clinical outcomes of AHF.

METHODSThis prospective, multicenter, observational study consecutively enrolled 3335 AHF patients who visited 14 EDs in Beijing from January 1, 2011, to September 23, 2012. Baseline data on characteristics and management were collected in the EDs. Follow-up data on death and readmissions were collected until November 31, 2013, with a response rate of 92.80%. The data were reported as median (interquartile range) for the continuous variables, or as number (percentage) for the categorical variables.

RESULTSThe median age of the enrolled patients was 71 (58-79) years, and 46.84% were women. In patients with AHF, coronary heart disease (43.27%) was the most common etiology, and myocardium ischemia (30.22%) was the main precipitant. Most of the patients in the ED received intravenous treatments, including diuretics (79.28%) and vasodilators (74.90%). Fewer patients in the ED received neurohormonal antagonists, and 25.94%, 31.12%, and 33.73% of patients received angiotensin converting enzyme inhibitors/angiotensin receptor blockers, beta-blockers, and spironolactone, respectively. The proportions of patients who were admitted, discharged, left against medical advice, and died were 55.53%, 33.58%, 7.08%, and 3.81%, respectively. All-cause mortalities at 30 days and 1 year were 15.30% and 32.27%, respectively.

CONCLUSIONSSubstantial details on characteristics and ED management of AHF were investigated. ABSTRACT

Objective @# To evaluate the tensile bonding strength (TBS) and antibacterial properties when an orthodontic adhesive was added with nanohydroxyapatite (nHAp). @*Methods@#Light cure orthodontic adhesive (Grengloo) was blended with 2% TiO2 containing nHAp nanoparticles by 0%、10%、20%、30% (w/w), while the control was not blended with nHAp. Brackets were bonded to extracted premolars by these new adhesives. TBS of 5groups were determined, and the adhesive remnant index (ARI) scores were assessed. Composite discs specimen were prepared, incubated with bacterial suspension for 48 h, and tested for antibacterial properties. @*Results @#No significant difference was found in ARI. The colony unit counts and lactate productionof the groups containing 2%TiO2 were significantly reduced. The colony unit counts and lactate production were without relationship with nHAp. The tensile bonding strength drastically decreased when containing more than 10% nHAp. @* Conclusin@#nHAp might not enhance the antibacterial effects of Grengloo.

ABSTRACT

Objective To explore the mechanisms of individual, social and environmental factors influencing physical activity and the relationships among the factors in patients with type 2 diabetes. Methods A survey was conducted from January to July 2015 among 225 patients with type 2 diabetes using convenience sampling. The questionnaire included 9- item Patient Health Questionnaire, the International Physical Activity Questionnaire,Perceived Social Support Scale, Chinese Walkable Environment Scale, Bandura′s Exercise Self- efficacy Scale. Results The effective questionnaire recovery rate was 98.7%(222/225). Exercise self- efficacy was a mediator of factors influencing physical activity. Exercise self- efficacy and physical activity were positively correlated (β=0.24, P<0.05). Walkable environment only affected physical activity indirectly medicated by exercise self- efficacy (β=-0.26, P<0.05). Perceived social support directly associated with physical activity (β=0.14, P<0.05). Depression not only affected physical activity directly (β=-0.28, P<0.05), but also associated with physical activity indirectly medicated by exercise self-efficacy (β=-0.32, P<0.05). Conclusions Interventions should be taken to increase the perceived social support, improve depression and walkable environment, especially to enhance exercise self- efficacy, so as to improve the physical activity level of patients with type 2 diabetes.

ABSTRACT

OBJECTIVE:To explore how to establish good procedure, system and mode of drug management, dispensing and application,in order to provide reference for risk management and rational use of rabies vaccine. METHODS:The use of rabies vaccine and storage temperature monitoring in our hospital during 2013-2014 were analyzed statistically, and risk management and use of rabies vaccine in our hospital were analyzed, and management measures and attentions were put forward. RESULTS:The amount of vaccinum rabiel (Vero cell) and human rabies immunoglobulin in our hospital were increased in 2014,compared to 2013. The position labeled with #1 in storehouse and that of labeled with #1 and #2 in dispensing store could meet the storage condi-tion of rabies vaccine. Risk management could be carried out in fields of drug requisition, cold chain management, drug manage-ment and drug dispensing. ADR should be paid attention. CONCLUSIONS:It is of significance to develop risk management and ra-tional medication guidance of rabies vaccine.
